A novel transmission electron microscopic method to study bacterial interaction

The author studied the effects of bacterial invasion on biofilm structure of another bacterium. Myxococcus xanthus and Bacillus subtilis were grown on 1.5% CYE agar. The cultures were incubated at 31° C for 3-4 days to give the colonies time to grow to a size of a few centimeters in diameter. M.xanthus colonies were plated next to B.subtilis colonies, by placing small drops of liquid cultures on the agar, so when the colonies grew they approached one another. Along with WT strains, EPS(-) strains were also prepared to see how the lack of EPS affected the interactions of the colonies. After incubation, the bacteria were prepared for TEM and viewed at several magnifications.
